Nowadays, 1,2-propanediol(propylene glycol) is a precious chemical that is used as UPR,
antifreeze, liquid detergent etc[1]. In general, a method derived from petroleum has been
used for producing 1,2-PD. But, it exists toxic coproducts, treatment of waste water[2]. In
our research, we would like to produce 1,2-PD from renewable sources, optimistically.
Methylglyoxal synthase gene(mgs) and glycerol dehydrogenase gene(gldA) obtained from
Escherichia coli K-12 was cloned in pYES2/CT and pYES3/CT, separately. The mgs gene
is concerned with changing from dihydroxyacetone phosphate(DHAP) to methylglyoxal
and gldA gene is concerned with changing from acetol to 1,2-PD[3]. After cloning, transformation
was accomplished into Saccharomyces cerevisiae. pYES2/CT vector produce URA3 and pYES3/CT vector produce TRP1. So, SDC(Ura-, Trp-) media was used for selecting transformants. Because vectors have the GAL1 promoter, galactose was added for cultivation in YPD media. After cell culture, 1,2-PD production was analyzed by HPLC (flow rate 0.5ml/min, oven temperature 40℃).
