Proteomic Approaches to Identify Constitutive Promoter Genes in Leuconostoc mesenteroides

Development of gene expression system for Leuconostoc mesenteroides, a GRAS microorganism found in Kimchi, armed with stable vector and strong promoter/terminator sequences might be potent for production of valuable materials. Strong constitutive promoter genes were identified by analyzing protein expression patterns for Leu. mesenteroides cells grown in glucose and sucrose. Genes encoding pyruvate kinase, ribulokinase, mannitol dehydrogenase and DNA-binding response regulator (Rec-WHTH domain) were constitutively
expressed in both carbon sources. This study introduces a novel approach to isolate strong promoter genes that can be used to construct food-grade expression system for Leu. mesenteroides.
