High-throughput Screening of Antimicrobial Peptides by ILAC(Immobilized-Lipid Affinity Capturing) Method and LC-MS Analysis

Our purpose on this study is high-throughput screening of antimicrobial peptides(AMPs) by ILAC method and LC-MS analysis. ILAC is the method that immobilizes phospholipid on magnetic nanoparticles and then captures AMPs by immobilized lipid affinity. AMPs act on phospholipid of plasma membrane to kill competitive microorganisms. We isolated a Lactobacillus curvatus and two strains of Lactobacillus sakei having antifungal activity against Candida albicans. Antimicrobial peptides were screened and isolated from three Lactobacillus strains, using
colorimetric assay and ILAC method. The molecular masses of AMPs were determined by ESI mass spectrometry. Antimicrobial spectrum of AMPs was tested using top agar plate or spot-on-lawn method against Gram-positive and Gram-negative bacteria.This present study is high-throughput screening of antimicrobial peptides from L. sakei and L. curvatus that act on phospholipid membrane disruption. And it is further expected to support the construction of AMP
database.
