Cytoprotective Effect of Hispidin Against Oxidative Cell Damage on Rat Insulin Producing Cells

Reactive oxygen species(ROS) have been shown to damage tissues. Thus, this demage has been considered as a potential cause for dysfunction of various organs in body. Some mushrooms, including Phellinus linteus and Inonotus hispidus etc., commonly produce a bundle of yellow antioxidant pigments that are composed of hispidin derivatives and polyphenols. Hispidin, metabolite of Phellinus linteus, is a potent antioxidant and inhibitor of protein kinase C(PKC).
This work has investigated the cytoprotective effect of hispidin against oxidative cell damage on rat insulin producing cells. As a results of chemical analysis, hispidin was found to scavenge 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ROS, hydrogen peroxide and superoxide radical, as well as other well-known antioxidants. In addition, we showed the hispidin reduced ROS and apoptosis of RINm5F induced oxidative stress. These results suggested that hispidin
protected cells against oxidative damage by scavenging produced ROS.
