Identification of biologically active materials in mud

Mud is widely known as an effective agent for skin care and already used in the cosmetic industry. Crude mud has been known to contain mineral constituents such as bentonite and germanium. However, the biologically active materials in mud have not yet been fully determined. To explore the biologically active ingredients in mud, we performed protease assays using casein as a substrate. The mud samples were extracted several kind of solvent, such as distilled water, Methanol (100%), Ethanol (100%) and Acetone(100%) for 24 hrs at room temperature. The mixtures were then filtrated and concentrated to dryness using vacuum evaporator. Cell tests such as melanin assay and cell viability assay were also performed. Free radical scavenging activity was determined. These results suggest that protease activity might play a role in enhancing the positive skin care effects attributed to mud.
