Agents causing depigmentation in human skin and its mechanism involved in melanogenesis pathway have been demonstrated. Their mechanism of action cover (i)tyrosinase inhibition, maturation and enhancement of its degradation, (ii)Mitf inhibition, (iii)downregulation of MC1R activity (iv)interference with melanosome maturation and transfer, (v)melanocyte loss, desquamation and chemical peeling. We had previously reported that skin whitening agent was isolated from methanol extract of LASA-B29 and has strong whitening activity in B16 melanoma cells. Methanol extract of LASA-B29 inhibited 43.7% melanin synthesis of B16 melanoma cells and exhibited low cytotoxicity (8.1%) at concentration 100 ug/ml. In this report, we demonstrated its chemical structure and its mechanism on the cell based. In elanogenesis, MITF activates the expression of melanocyte specific markers and promotes the tyrosinase and related protein which are major melanogenic protein. Thus in order to determine whether purified compound represses Mitf activity and following the tyrosinase and related protein.
