Effects of Temperatures in Transgenic Oryza sativa L. Cell Cultures Producing hCTLA4Ig

The optimal temperatures for cell culture were varied with cell types and culture stage. Therefore, two-stage process switching temperature was applied to produce hCTLA4Ig in transgenic rice suspension cell cultures. Combinations of 28℃ and 25℃ were used for growth and production: (A) 28℃→28℃; (B) 2 8℃→25℃; (C) 25℃→25; (D) 25℃→28℃. The maximum level of hCTLA4Ig was achieved in the case (A) on day 6. When hCTLA4Ig production was high in
each combination of temperature, highly expressed intracellular proteomes were selected and analyzed for the relationship with production. Except for the case (A), three proteomes were found to be increased with productivity. These results could provide useful information for the improvement of cell cultures and target protein production.
