Bioconversion of ginseng root extract to ginsenoside compound K by β-galactosidase form Sulfolobus solfataricus

Ginsenoside compound K, as a component of Panax ginseng, has been known for the anti-inflammatory, anti-proliferative, and anti-carcinogenic activities. Ginsenosides Rb1, Rb2, Rd of ginseng root extract were converted into pharmaceutically active minor ginsenoside compound K using β-glucosidase from Sulfolobus solfataricus. The enzyme activity to produce ginsenoside compound K was optimal at pH 5.5, 90 ℃, and 20 U/ml, and the enzyme was stable below 85
℃. Under stable conditions of pH 5.5, 85 ℃, and 20 U/㎖, the β-galactosidase produced 1.63 g/l of compound K from ginseng root extract (Rb1 1.9 g/l, Rb2 0.52 g/l, Rd 0.23 g/l) at reaction time of 12 h.
