Silica-Armored Alginate Beads for Encapsulation of Protein Transcription/Translation Machinery: Towards Artificial Cells

Alginate has been a most popular encapsulation material for proteins and cells with their conserved activity.1 In this study, the cell-free transcriptional/translational protein synthesis machinery was encapsulated in the silica sol-gel coated calcium alginate beads in order to prepare individual vehicles of gene expression. The productivity of prepared vehicles were
investigated with the beads incorporated with plasmid DNAs encoded to produce the enhanced green fluorescence protein (EGFP), and they successfully expressed the target protein. The beads containing 1.5 to 2.0 wt.% of alginate exhibited the maximum fluorescent intensity. In addition, the silica-armored vehicles continuously expressed EGFPs for 50 hours and endured the expression condition for more than 100 hours in comparison with 20 hours of production
and 40 hours of endurance of unarmored beads. The result of this work might provide a possible application towards the artificial cell producing functional protein.
