Transgenic rice cells as a production system for recombinant proteins have a problem of a slow rate of growth in AA medium1,2,3). To improve the growth rate of transgenic rice cells, we have investigated ;the effects of initial sucrose concentrations, the aggregated size of rice cells, and peptide mixtures such as casein hydrolysate on the cell growth in AA medium. In 60 g/L of initial
sucrose and above, increase of cell mass was observed in contrast to control. However, the time required for obtaining maximum cell density was not reduced compared to that in control. When the aggregated cells were sieved with 20-mesh screen, 11 g/L of dry cell weight in the sieved cells could be achieved at 12 days, whereas the non-sieved cells reached 10 g/L of dry cell weight at 14 days. The sieved cells could reach to maximum cell density 2 days earlier
than the non-sieved cells. Using peptide mixtures, such as peptone, casitone, and casein hydroylsate, time required for maximum cell density could be significantly moved from 12 days to 6 days. During the exponential phase of the growth, 0.40 d-1 of maximum specific growth rate was obtained in AA medium using 2% casein hydrolysate.
