Polyunsaturated fatty acids (PUFAs) are essential components of human health. Linoleic acid (LA) and α-linolenic acid (ALA) are precursor fatty acids that are elongated to form long chain polyunsaturated fatty acids (LCPUFAs) like arachidonic acid (AA), eicosapentanoic acid (EPA) and docosahexaenoic acid (DHA). During the last decade, desaturase and elongase genes responsible for the biosynthesis of PUFAs have been cloned from various organisms. A TaELO
elongase gene was isolated from T. aureum ATCC 34304 contains a high level of PUFAs. A TaELO gene was cloned into the methylotrophic yeast P. pastoris GS115. In this study we confirmed optimum methanol induction time and substrate reaction time conditions of C20-Δ9, C20-Δ6 elongation. Consequently, optimum condition of C20-Δ9 elongation was 48 hr of methanol induction and substrate reaction and C20-Δ6 elongation was 36 hr of methanol induction and substrate reaction.
