Optimization of the medium composition for Bacillus coagulans using response surface methodology

A three level Box-Behnken factorial design was employed combining with response surface methodology (RSM) to optimize the medium composition for Bacillus coagulans culture.1) Bacillus coagulans is an atypical strain for lactic acid and industrial production of lactate.2) The cost for the production of higher cell mass can be reduced by optimization of inexpensive complex medium components. Three MRS (deMan Rogosa Sharpe) medium components (e.g.
yeast extract, malt extract and glucose) were selected to be optimized. The estimated optimum concentrations of yeast extract, malt extract and glucose was 16, 8.4 and 33.7 g/L, respectively. The amount of produced Bacillus coagulans at optimum composition was 1.7 gDCW/L. More than 10% of higher cell mass was obtained comparing with conventionally used medium composition.
