Natural -carotene has been used as a functional β cosmetic agent because chemical synthetic one has been severely regulated in cosmetic industry.
Although anti-wrinkle properties of β-carotene has reported, no experimental evidences and/or optimal dosage were not revealed. In this presentation, we described the extraction of beta-carotene from metabolically engineered E.coli, purification by crystallization, and evaluation of anti-wrinkle efficacy of purified beta-carotene. 223 g/L of wet cells was harvested from 2.5-L fed-batch culture broth. After cell disruption by sonication, dehydration using acetone, extraction
with isobutyl acetate at 60oC for 10 min. and crystallization of cell extracts at 4oC for 12 h. 633 mg of β-carotene was obtained with a purity of 93% as measured by HPLC. Isolated beta-carotene was evaluated by performing collagen synthesis assays using human normal fibroblast cells. 3.5 μM of beta-carotene was found to be optimal concentration at which 1.4-fold higher amount of collagen was synthesized than that in absence of beta-carotene. This indicates
that beta-carotene derived from recombinant E.coli can be used in functional cosmetics as anti-wrinkle agent.
