Process Development of Solvent Extraction Method for Melanin and Atopy Activity in Placenta Cosmetics

The placenta extract was commercially used for the health foods, cosmetics and pharmaceuticals. In order to isolate the useful materials from raw pig placenta, the hydrolysis or enzymatic methods was generally used. In the present study, the solvent extraction method was newly developed to obtain more valuable compounds such as proteins, amino acid, etc. The solvent extraction methods was investigated by introducing ethanol, hexane, ethyl acetate and butanol. The sample placenta was prepared in the Tamra hog farm (Jeju, Korea). The placenta was extracted with 80% ethanol and 50g placenta powder for 24 hours.
This extraction powder was partitioned by hexane, ethyl acetate and butanol(12H, 25℃), respectively. In order to compared, the placenta extract was used as the methods of hydrolysis and enzyme resolution. As a control experiment, placenta extraction was performed by 5 times of homogenization and twice of sonication after the PBS treatment. The melanin activity was analyzed at 400 nm using an ELISA reader. Atopy activity was also measured in a Peltier Thermal Cycler (PTC-100, MJ research, USA). This comparative study was clearly showed that the extract of ethyl acetate layer was more efficient than different methods in the melanin and atopy activity.
