Skin whitening effect and regulation of proteins related to melanogenesis by Angelica gigas extract

Skin whitening effect of Angelica gigas extract has been analyzed. Angelica gigas extract was obtained at pressure of 300 bar and temperature of 60℃ by supercritical carbon dioxide extraction system. To decide the proper treatment concentration of extract, MTT assay was performed with B16-F1 Melanoma.
Angelica gigas extract was showed cytotoxicity at over 10 mg/L concentration. Tyrosinase IC50 value of Angelica gigas extract was 49.89 mg/L, the same as arbutin (49.52 mg/L) which is well known skin whitening material.
Melanogenesis inhibitory effect with melanoma was showed high activity at concentrations of over 0.5 mg/L. Regulation of proteins related to melanogenesis by Angelica gigas extract was investigated using proteome analysis. The proteome of malanoma with or without Angelica gigas extract were profiling by 2D-PAGE. In conclusion, it was shown that many protein factors related to melanogenesis were down-regulated by Angelica gigas extract1,2).
