Microcarriers have been used for suspension culture of anchorage-dependent cells.1) For scale-up or cell recovery, cell detachment by trypsinization is required. However, the trypsinization process requires labor and addition steps and may have detrimental effects on structure and functions of the cultured cells. In this study, we developed microcarriers from which cultured cells can be detached without trypsinization. Thermo-sensitive microcarriers were fabricated
by incorporating poly-N-isopropylacrylamide (pNIPAAm) on Cytodex-3. The pNIPAAm grafted Cytodex-3 (pNIPAAm-Cytodex-3) microcarrier allowed bone marrow-derived mesenchymal stem cells (BMMSC) to adhere, spread, and grow successfully on the surface of the microcarriers. After the cell culture, most of the cells were detached from the thermoresponsive Cytodex-3-pNIPAAm by simply lowering the temperature to 32 ℃. Cell detachment from the
pNIPAAm-Cytodex-3 microcarriers by temperature drop significantly reduced cell damage and apoptosis compared to trypsinization. pNIPAAm-Cytodex-3 microcarrier would make cell culture scale-up process more simple, easy, and less laborious. This procedure could be also valuable to the variety of potential fields, demanding large amount of cells without damage, such as cell therapy, and tissue engineering.
