N-Nitrosamines, which have long received attention in the fields of food hygiene and environmental health, have been detected in several drug substances and drug products. Nitrosamine impurities were continuously detected in a number of pharmaceuticals such as Ranitidine, Nizatidine, and Metformin, starting with angiotensin receptor blockers such as valsartan. As a result, domestic and foreign regulatory agencies took measures to inspect the collection, discontinue manufacturing, and recall the drug, and the controversy over the safety of the drug has been raised for the past three years. In this paper, various official compendial and academic papers reported on the analysis method of Nitrosamine impurities among pharmaceuticals are summarized by classifying them into Sartans, Ranitidine·Nizatidine, and Metformin drugs. The advantages and disadvantages of liquid chromatographymass spectrometry (LC-MS) and gas chromatography-mass spectrometry (GC-MS), which are mainly used, were reviewed. In addition, the pre-treatment method for the analysis of Nitrosamines among pharmaceuticals, the cause of its formation, and precautions are also summarized.

식품 위생과 환경 보건 분야에서 오랫동안 주목을 받아온 니트로사민류 화합물이 일부 원료의약품과 완제의 약품에서 검출되면서 전세계적인 관심의 대상이 되었다. 발사르탄을 비롯한 안지오텐신 수용체 차단제 계열의 의약 품을 시작으로 라니티딘, 니자티딘, 메트포르민 등 다수의 의약품에서 니트로사민류 불순물이 계속해서 검출되는 사 례가 발생하였고, 이에 국내외 규제기관에서는 해당 의약품의 수거 검사와 생산 중지, 회수 조치를 취하는 등 의약품 안전성에 대한 논란이 지난 3년간 계속하여 제기되었다. 본 논문에서는 사르탄류, 라니티딘·니자티딘, 메트포르민 의 약품으로 분류하여 의약품 중 니트로사민류 불순물의 분석법에 대하여 보고되었던 각종 공정서와 학술논문을 정리하 였다. 주로 사용되고 있는 액체크로마토그래피-질량분석법(LC-MS)과 기체크로마토그래피-질량분석법(GC-MS)의 장 단점과 고려할 사항에 대하여 고찰하였고, 의약품 중 니트로사민류 분석을 위한 전처리 방법과 생성 원인 및 주의할 사항에 대해서도 정리하였다.
