Characterization of Size and Internal Material Content for the Vesicle by Asymmetrical Flow Field-flow Fractionation (AF4) and UV/Vis Spectrophotometer

The vesicle can storage and transport materials and have the same structure as the human body's cell membrane, thus attracting attention in various industries such as drug delivery systems (DDS) and transdermal delivery systems (TDS). The vesicles' size and stability are important factors in DDS and TDS as they affect the amount of the material and the time span over which the material is released. Therefore, it is important to make vesicles that are stable in size. In this work, the vesicle's size was optimized, and an analysis was performed to determine the material contained within the vesicle. To optimize the vesicles' size, the concentration of lipids and the extrusion flow rate, which can affect the size of the vesicles, were optimized, and results were obtained as < 0.4% and 0.3 mL/min, respectively. Also, we confirmed that 2.6 μM of calcein was contained in the calcein containing vesicle by the internal standard method. From these results, AF4 can be a useful tool for characterizing vesicles' size and size distribution. It also provides its applicability as a separating and preparing method to obtain the desired size range vesicles.
