The aim of this study was to identify styrylpyrone-class polyphenols with anti-inflammation and anticancer from fruiting body of Phellinus linteus KACC93057P that was artificially cultivated on oak logs. The fruiting body was extracted using methanol and partitioned using hexane, chloroform, ethyl acetate, and butanol, successively. The ethyl acetate-soluble layer was further separated using silica gel and Sephadex LH-20 column chromatography and preparative HPLC to provide three polyphenolic compounds. These compounds were identified as hispidin, hypholomine B, and davallialactone based on mass and NMR spectroscopic methods. The hypholomine B and davallialactone showed proliferative activity over 50% in LPS (lipopolysaccharide) -activated RAW 264.7 cell at 250 μg/mL, while hispidin exhibited an inhibitory effect on the cell viability of 30%. Hypholomine B, and davallialactone inhibited NO production of LPS activated RAW 264.7 cell by 25-95% in at the concentrations (15-250 μg/mL), showing their effects on anti-inflammation. The compounds inhibited cytokines, IL-6 and TNF- production of 83-93% in the cell at 100 μg/mL and further they showed inhibition rate of 42-50% in HCT116 human colon cancer cells at concentration of each 250 μg/mL, showing morphological alternations with cell shrinkage and irregularity of shape in the cancer cells.
