Different Regulation of Plasminogen Activators and Their Inhibitor by Progesterone and 17β-Estradiol in Porcine Uterine Epithelial Cells

This present study was conducted to investigate different regulatory pattern of mRNA expression and activity of two-type plasminogen activators (PAs; urokinase-type, uPA; tissue-type. tPA), uPA specific receptor (uPAR), and type-1 PA inhibitor (PAI-1) by different concentration and exposure time of progesterone (P4) and 17β-estradiol (E2) in porcine uterine epithelial cells. Cultured epithelial cells were incubated with different concentrations of P4 or E2 (0, 0.1, 1, 5, 10, and 20 ng/mL) for 5 min or 24 h, respectively. In results, 24 h treatment of P4 or E2, and E2 treatment for 5 min did not affect to mRNA expression of uPA, tPA, uPAR, and PAI-1. In P4 treatment groups for 5min, the mRNA expression of uPA, uPAR and PAI-1 were increased in 20 ng/mL P4 groups compared with 0.1 ng/mL P4 treatment groups (p<0.05), whereas it is no different with control group (0 ng/mL). On the other hand, 10 and 20 ng/mL P4 treatments enhanced the mRNA expression of tPA compared with 0 ng/mL treatment in epithelial cells (p<0.05). When these cells were exposed to P4 or E2 for 5 min, only 0.1 ng/mL P4 increased PAs activity (p<0.05). Although mRNA expression of PAs were not influenced by exposure of sex hormone for 24 h, PAs activity was increased by 10 and 20 ng/mL treatments in both P4 and E2 (p<0.05). In conclusion, short term treatment of P4 upregulated uPA, tPA, uPAR. and PAI-1 mRNA expression, but E2 did not affect in both of short- and long-term treatment. Interestingly, PAs activity was increased by long term treatment of P4 and E2. These results show that mRNA expression of PAs are differently regulated by different concentration and exposure time of sex hormones, however, pattern of PAs activity is different with mRNA expression. Therefore, more studies regarding with regulatory mechanism of PA system are needed.
