Cordycepin is one of the most crucial bioactive compounds produced by Cordyceps militaris and has exhibited antitumor activity in various cancers. In the present work, the response surface methodology (RSM) was used to determine optimum conditions for the extraction of cordycepin from C. militaris fruiting body. Extraction conditions, such as extraction solvent and extraction time were optimized using RSM. Analysis of variance (ANOVA) was applied and the significant effect of the factors and their interactions were tested at 95% confidence interval. Extraction solvent composition and time affected the extraction of cordycepin from C. militaris and the maximum amount of cordycepin was obtained when the samples were extracted using 50% methanol for 4 h. Furthermore, Diaion HP-20 column was successively used to isolate and purify cordycepin from C. militaris. Water crude extract of C. militaris was fractionated using Diaion HP-20 composed of water-methanol (W:M) and fraction (W:M 60:40) yielded 63.8% of cordycepin, and the fraction (W:M 80:20) produced 33.7 % consecutively, whereas the methanolic crude extract yielded 57 % and 38.6 % respectively. The purity of the prepared cordycepin was 93.2 % in one-step separation according to the high performance liquid chromatography (HPLC) analysis. The preparation method is simple, highly efficient, energy saving, and has been demonstrated to be effective for large preparations of cordycepin from crude extract with high purity and low equipment and operating costs.
