Specific PCR primer set for detection of Trichoderma spp. causing green mold disease during mushroom cultivation

We have developed the primer set for detection of Trichoderma species, which cause green mold disease in Pleurotus eryngii and Pleurotus ostreatus in the mushroom farms. Based on the sequences of the internal transcribed spacer (ITS) region in T. harzianum, T. longibrachiatum, T. atroviride, T. koningii, T. cf. virens, T. harzianum, T. pleuroticola, T. pleuotum and T. citrinoviride, four primers composed of two forward, TDP F1 and TDP F2, and two reverse, TDP R1 and TDP R2, primers were designed. Among the four possible combinations of these primers, PCR with TDP F1 and TDP R2 primer set detected all Trichoderma species of previously mentioned. When the performed PCR with this primer set and mushrooms of P. eryngii, P. ostreatus and Grifola frondosa, the expected fragment was not amplified. This study will be useful for Trichoderma species, which causes green mold disease during mushroom cultivation, detection in contaminated cultures of mushrooms such as P. eryngii, P. ostreatus and G. frondosa.
