Roles of Dia-related Formins in Spindle Formation and Oocyte Maturation

Dynamic actin reorganization is essential factor for the maturation of mammalian oocyte maturation. Actin nucleator, includig Arp2/3 complex and formins, which initiates actin filaments, are key regulators for these processes. In mammalian, 15 formin family of proteins are exists and one of them known as formin-2, has been known to play crucial roles in spindle migration of oocyte. but roles of other formin family proteins in oocyte maturations are unknown. To shed light on functions of formin family proteins in oocyte maturation, we utilized SMIFH2, recently developed small molecule inhibitors of formin family proteins to check the effect of formin family proteins in oocyte maturation. Treatment of 500 μM of SMIFH2 completely block polar body extrusion of mouse oocyte. Detailed investigation showed that treatment of SMIFH2 in GV stage oocyte severely impair formation of meiotic spindle, revealed the novel roles of formin family proteins in spindle formations. Using RNAi against mammalian Diaphanous-related (mDia) formins, mDia2 and mDia3, we are investigating the roles of Diaphanous-related formins in meiotic spindle assembly and oocyte maturation.
