Hyperpigmentation diseases especially melisma and lentigines are major psychological disease in most of the societies. The obsession for the clean, fare and healthy skin is one of the basic instincts to every individual. To such diseases, a number of melanogenesis inhibitors have been screened. They were effective but brings to many complications as their side effects. Pink eye dilution protein (P-protein) is a melanogenogenic protein and plays a critical role in cellular melanogenesis. The present study is aimed to develop an alternative molecular approach using P-protein small interfering RNA (siRNA) to control melanogenesis in the melan-a melanocyte. It has been observed the both P-protien and mRNA level were significantly lowered by the siRNA treatment. P-protein siRNA also suppressed melanin synthesis without any cytotoxicity in the melan-a melanocyte cells. These results suggest that molecular approaches using siRNA targeting P-protein may a novel approach for the control of the melanogenesis in a safer way.
