A number of particular attention have been placed into the colorimetric assays using gold nanoparticles (AuNPs) in the field of clinical and environmental diagnosis,[1][2] yet there are only a few attempts to detect protein phosphorylation, due to low selectivity and affinity. Here we describe a rapid visualization of phosphorylated peptides based on zinc-coordinated self-assembly of AuNPs. While the phosphorylated peptide with hexahistidine at a single end was added to the carboxyl AuNPs, a strong association between the carboxyl group of AuNPs and two regions (hexahistidine and phosphate group) of the peptide was observed in the presence of zinc ion, which was followed by the color change (from the red to violet) by the self-assembly of the AuNPs. By contrast, the control unphosphorylated peptide, the hexahistidine-free phosphorylated peptide and the addition of other metal ions, such as nickel, copper, cobalt ions, did not change the color of AuNP solution. When the self-assembly of the AuNPs was quantified by the extinction ratio (E520/E700), the concentration of phosphorylated peptide was easily determinated. In this regard, we anticipate that the developed method will find the application to analyze the activities of various protein kinases with high selectivity and rapidity.
