Human myoglobin can be used as an early marker indicating the onset of Acute Myocardial Infaction (AMI). A gene for the single chain variable fragment (scFv) of an anti－human myoglobin antibody was constructed, and cloned in plg20m－sc1 vector with a Protein A tag that specifically binds to IgG. The scFv fused with the Protein A tag was produced in E. coli through IPTG induction at 30℃. The recombinant scFv was purified through an affinity chromatography using IgG sepharose resin. There were two protein bands in the elution fraction. Western blot analysis using an anti－His tag antibody revealed that a 40 kDa protein was confirmed as the Protein A fused scFv was purified using gel permeation chromatography. The fused scFv bound to human myoglobin in a concentration dependent manner.
