Human myoglobin can be used as an early marker indicating the onset of Acute Myocardial Infaction (AMI). A gene for the single chain variable fragment (scFv) of an anti-human myoglobin antibody was constructed, and cloned in plg20m-sc1 vector with a Protein A tag that specifically binds to IgG. The scFv fused with the Protein A tag was produced in E. coli through IPTG induction at 30℃. The recombinant scFv was purified through an affinity chromatography using IgG sepharose resin. There were two protein bands in the elution fraction. Western blot analysis using an anti-His tag antibody revealed that a 40 kDa protein was confirmed as the Protein A fused scFv was purified using gel permeation chromatography. The fused scFv bound to human myoglobin in a concentration dependent manner.
