A Novel Gene Expression System for the Regulation of Epitope Density on the Protein Nanoparticle Surface.

Protein nanoparticle(PNP) are being used in many fields such as vaccine, diagnosis and treatment, which is done by locating target epitopes on their surface. Various kinds of epitopes can be displayed on the surface of the PNP by co-expression. The density of the epitopes on the PNP can be regulated by using sigma 32 dependent promoter. Human ferritin heavy-chain(hFTN-H) and HBV capsid(HBcAg) are the most widely used stable PNP. We confirmed that fusion expression of target epitopes and the C-terms of hFTN-H and the tips of prominent surface spikes of HBcAg resulted in the successful expression. We inserted ‘sigma 32 dependent promoter’ into the expression vector for the reduced density of the specific target epitopes. As a result of co-expression, we found that the density was dramatically changed. In conclusion, the novel gene expression system promotes the efficient use of PNP by the regulation of their surface density.
