Collagen has been widely used tissue engineering biomaterials in the last two decades due to its mimicking properties of human protein. In the present study, we have isolated collagen from Thunnus obesus bone and used for bone graft substitute. Two different kinds of methods have been used to isolate the collagen; there are Acid Soluble Collagen (ASC) and Acid Soluble Enzyme Collagen (ASEC) methods. The isolated collagen has been characterized with Fourier Transform Infrared Spectroscopy (FT-IR), SDS-polyacrylamide gel electrophoresis (SDSPAGE), Optical Microscopy (OM) and Scanning Electron Microscopy (SEM). FT-IR results reveal that presence of pure collagen. SEM and OM results depicted that collagen is in the presence of nano fiber sponge like scaffolds. The isolated collagen scaffold is checked with pre osteoblast (MC3T3-E1) cell line. The in vitro results reveal that collagen scaffolds are highly biocompatible and non-toxic in nature. Herewith, we are suggesting that natural derived marine fish collagen scaffold will be excellent biomaterials for bone tissue engineering scaffold.
