Mesenchymal stem cells (MSCs) from umbilical cord (UC) can be used for regenerative medicines. Generally, enzymatic isolation method of the cells from UC was simple and safe than mechanical method. But detailed characterization of cells between two methods is not studied yet. In this study, we investigated the comparison of MSCs characteristics depending on their isolation methods for optimized stem cell isolation. The cells from two different methods were showed similar duplication rate, morphology and CFU-F assay through long term culture. Also, the protein expression of Col type I, HLA class I, CD73 and CD90 were showed same patterns by ICC. Gene expression profiling of UC-MSC analyzed by Human Whole Genome Array showed similar results according to isolation methods (>99%). Also, the differentiation potency of bone and adipocyte were not appreciably different in. These data showed that there is no different effect on the character of UC-MSCs according to their isolation methods. And the enzymatic isolation method of hUC-MSCs can be used to optimized and standardized method in cell therapy in the future.
