Establishment of Optimal Culture Conditions for Human Umbilical Cord Derived Mesenchymal Stem Cells(hUCMSCs) by Gene Expression Analysis

hUC-MSCs were important sources of regenerative medicine. And it should have the stable proliferation condition and it shouldn’t show alteration of gene expression pattern after culture. This study was performed for establishing of stable culture condition of hUC-MSCs for cell therapy. The hUC stromal cells derived from same donor were isolated by enzymatic dissociation method. After culture in different media, proliferation potency and alterative gene expression pattern were compared. Gene expression patterns were analyzed by RT-PCR and ICC. Morphology of cultured cells was similar except in MSC-qualified medium A. And medium A, C and D showed significantly higher proliferation ability than medium B. MSC specific markers were expressed positively for ADAM12, ALP, CD73, CD90, CD105 but negatively for CD31, CD34, HLA class II DR in medium A and D. But expression of HLA class II DR was changed in medium C. Our data suggest that medium D can be used for hUC-MSC culture. In conclusion, we can see the altered gene expression pattern by culture conditions. And we can establish the optimal culture condition of hUC-MSCs and it can use standardized method in cell therapy in the future.
