Biosilica based enzyme immobilization by silica forming peptide.

Silicon is the second most abundant element in the Earth’s crust. However, silicon does not find alone in nature. It always exist stable form as silica or silicates. Especially, silica has biocompatible characteristic and suitable for bio materials. Silica forms covalent bonding with specific chemicals. Therefore, silica is good material for organic-inorganic hybrid complex. Common manufacturing process of silica particle is based on extreme environment such as high/low pH, high temperature, and high pressure. We solved these problems by biosilicification which comes from diatom’s bio synthetic process for cell wall. At first, we found new SFP(silica forming peptide) which forms biosilica. Next, we proved protein display on the silica particle by GFP-SFP fusion protein. So, we fused SFP with cellulase which hydrolyzes cellulose and made biosilica immobilized enzyme. As a result, biosilica immobilized cellulase showed high thermo stability and maintained 50% activity until 10 times recycling. We expect enzyme immobilization method by SFP provide better condition to industrial applications.
