Expression of Recombinant Human Bone Morphogenetic Protein 2 (rhBMP2) in Silkworm Pupae and Cells.

The secreted bone morphogenetic protein 2(BMP2) is important role in the development of bone and cartilage. Recently, rhBMP-2 protein has become immensely popular because of the need for larger bone graft amounts required for this procedure especially in patients with poor and limited bone quality. Here, we examined the large-scale production and purification of BMP2 in silkworm pupae and silkworm cells. The human bone morphogenetic protein 2 (BMP2) was cloned with Hela cell library and efficiently expressed in silkworm pupae and silkworm cells using baculovirus expression system (BES). BES in silkworm factory had a clear band for rhBMP2, a homodimeric protein with an apparent molecular weight of about 52 kDa with carbohydrate.In this study, we describe a procedure for the expression and purification of human BMP2 from silkworm using BES. We believe that these results solve current problems of sufficient amounts and cost impeding routine clinical use of BMP2.
