Antibiotic regulatory gene, MarR-like regulator AbsC (SCO5405) of Streptomyces coelicolor was previously found to affect productions of antibiotics, actinorohdin (ACT) and undecylprodigiosin (RED).1 When AbsC deletion mutant was grown in minimal media containing glucose and alanine as a C and N source, the intracellular level of pyruvate and α-ketoglutarate decreased while the production of ACT and ammonium ion increased.2 To investigate such phenomena and characterize roles of AbsC, we performed metabolite profiling with TSQ LC-MS/MS. Metabolite profiling revealed accumulation of malate and oxaloacetate, but depletion of citrate, isocitrate, and α-ketoglutarate in AbsC deletion mutant. In addition, AbsC was either directly or indirectly repressing glyoxylate pathway and transdeamination process, confirmed by expression levels of TCA and glyoxylate cycle related genes and the two enzymes of transdeamination with Real Time PCR (qPCR). From the profiling and qPCR data, we could conclude that AbsC was one of rare regulators that controlled both primary and secondary metabolism.
