Homogentisate 1,2-dioxygenase catalyzes dioxygenation of homogentisate in tyrosin or phenyalanine catabolism1-2. The catalyst is a metal ion binding protein that is required especially iron as a cofactor3-4. The sequence identity of homogentisate 1,2-dioxygenase in Comamonas sp. strain P19 is closely matched with glyoxalase in GenBank database and the bifunctional activities for dioxygenase and glyoxalase I were observed by pH dependent manner. In this study, recombinant homogentisate 1,2-dioxygenase of strain P19 was purified and crystallized to determine the three-dimensional structure. The comparison of crystal structures of the enzyme under homogentisate 1,2-dioxygenase or glyoxalase I activity will give us a clue for elucidation of the bifunctional mechanisms.
