The efficient enrichment process coupled with random mutagenesis can be an effective approach for the improvement of microalgal strains to increase biodiesel production. In this study, we developed an screening process based on continuous cultivation system in a 2 L flat-plate photobioreactor to screen mutants which exhibit fast growth rate. Ethyl methanesulfonate (EMS) was used to induce mutation from oleaginous Chlorella vulgaris. Positive selection for mutants with high specific growth rate was achieved via feedback control between media pump and online measurement of culture density. Cells with low specific growth rate is washed out when their growth rate is lower than operating value of dilution. The method was validated by mix-cultivation of four different wild type microalgae, i.e. Chlorella vulgaris, Chlorella protothecoide, Chlorella zofingiensis, and Chlamydomonas reinhardtii. During cultivation, the lipid content of the mix-culture was analyzed with FACS to track the change of culture population.
