dagA as β-agarase degrades agarose into neoagarotetraose and neoagarohexaose as major products. Neoagarooligosaccharides including above two have a good potential as active ingredient in the pharmaceutical, food, and cosmetic industries. The major objective of this study was to develop an optimized medium for the production of dagA by employing Streptomyces lividans. Two very popular statistical methods of Plackett-Burman design (PBD) and the response surface method (RSM) with a central composite design (CCD) were employed to select key components and to optimize their concentrations, respectively. Among 7 nutrients tested, one carbon source of glucose, one nitrogen source of yeast extract, and MgCl2 · 6H2O were selected through PBD as medium components whose concentrations were to be optimized. Through the RSM experiment, the optimum concentrations of glucose 25 g/L, yeast extract 11 g/L and MgCl2 · 6H2O 5 g/L were identified. With the optimized medium composition, 7.42 unit/ml of dagA was produced in 3 days of cultivation.
