Ammonia-oxidizing archaea (AOA) are ubiquitously distributed and plays a pivotal role in the global nitrogen cycling. Although the demand for elucidating functional roles in natural habitats is being increased, their physiological and biochemical characteristics remain largely unresolved. AOA have been isolated from various habitats using ammonia as a sole energy source. Recent discoveries have further suggested that polar archaea have potential for utilizing urea as an energy source, based on deduced urease-utilizing genes (ureA, ureB and ureC). In this study, we confirmed successful cultivation of AOA with urea by immunostaining using archaea specific anti-AMO antibody. Furthermore, we first reported proteome of AOA grown on ammonia or urea as a sole energy source by using 2-dimensional gel electrophoresis and peptide mass fingerprinting. It is expected that our study will contribute to resolve physiological and ecological role of AOA in natural habitats.
