Improved Cell Viability of Lactobacillus crispatus KLB46 by Heat Adaptation

Lactobacilli have important roles in the human vagina that prevent vaginal infection by pathogenic organisms. In this study, we selected Lactobacillus crispatus KLB46 isolated from human vagina that showed high antimicrobial activity. Thermotolerant is important to pharmaceutical formulation using spray drying process. we confirmed heat adaptation for increasing thermotolerant of KLB46. Viable cell population of KLB46 at 60℃ for 20min was reduced 108-109 to 102-103. However adapted KLB46 at 52℃ for 15min showed high cell viability than non-adapted cells about 103 folds at 60℃ for 20min. To optimize the adaptation temperature, we conducted adaptation at different temperature(42℃, 47℃ and 52℃) and the highest cell viability showed at 52℃. To characterize the difference in cell surface property, we conducted hexadecane-water adhesion assay. Cell surface hydrophobicity is an important factor for adhesion to human vagina. The results showed that cell surface of the adapted cell was more hydrophobic than the non-adapted cell.
