Cloning of Genes Encoding Acetolactate Decarboxylase, 2,3-Butanediol Dehydrogenase from Various Bacterial Strains for the Production of 2,3-Butanediol

2,3-Butanediol can be utilized in a variety of industrial products such as chemical feedstock, antifreeze agent, industrial solvent methyl ethyl ketone, rubber production, diacetyl. 2,3-Butanediol can be biologically produced from glycerol using microorganisms. Acetolactate synthase, acetolactate decarboxylase and 2,3-butanediol dehydrogenaseare three key enzymes for the biological production of 2,3-butanediol. In order to increase the production of 2,3-butanediol, many kinds of acetolactate decarboxylase, 2,3-butanediol dehydrogenase genes were isolated from various bacterial strains such as Klebsiella pneumoniae, Serratia marcescens, Bacillus subtilis and so on. These genes were identified by analysis and comparison of the nucleotide sequences, which were cloned into Escherichia coli expression vector to identify its function. acetolactate decarboxylase and 2,3-butanaediol dehydrogenase genes was cloned into expression vector pACYCDuet-1. Then, cloned acetolactate decarboxylase and 2,3-butanaediol dehydrogenase genes were expressed by IPTG induction, and protein expression was confirmed by SDSPAGE. After culturing the microorganisms, the culture supernatant was analyzed by HPLC. Finally we could select some recombinants that showed a significant increase in production of 2,3-butanediol.
