4-hydroxybenzoic acid (HBA) is a precursor of cis,cis-muconic acid which is applied to various industrial fields such as synthesizing nylon and polyurethane. Corynebacterium glutamicum, which is a gram-positive soil bacterium and has ability of aromatic degradation and assimilation, produces a large amount of chorismic acid which is the precursor of 4-HBA, so it is one of the promising microorganisms for muconic acid production. In this study, the metabolic flux network of Corynebacterium glutamicum was constructed by diverse metabolic flux analysis (MFA) tools. Then based on the MFA results, several mutants of Corynebacterium glutamicum were made. The mutants were engineered to produce large amounts of 4-HBA, and the experimental data were compared with the MFA results. For increasing and analyzing the 4-HBA production, MFA which is a powerful tool established on the stoichiometric model was used. By calculating the intra-cellular fluxes it was possible to understand the biosynthesis of 4-HBA in C. glutamicum. In conclusion, the recombinant strain over-expressing the ubiC gene showed the highest amount of 4-HBA, 7.9 g/L.
