The aim of this study was to develop recombinant Saccharomyces cerevisiae (S. cerevisiae) with improved fatty acids through genes involved in fatty acid synthesis elongation cycle. The recombinant strains included fabF from S. cerevisiae YPH500 and fabG, fabZ, fabK, and EC.3.1.2.14 from Streptococcus pyogenes MGAS 10270; these strains were developed using pESC-HIS, pESC-LEU, and pESC-TRP expression vectors, respectively. Production of fatty acid (C16) from SGJY01 (containing fabF) increased about 1.8-fold as compared to the wild-type strain. The fatty acid composition in the S. cerevisiae membrane varied significantly in response to environmental stresses, such as changes in temperature. To confirm the effect of temperature in fatty acid composition, the recombinants and wild-type were cultured at different temperatures (30, 20 and 10oC). As the culture temperature was lowered, the unsaturated fatty acids (C16:1, C18:1) of SGJY01 were increased up to 2.4-fold as compared to the wild-type. Of particular interest is that the proportion of unsaturated fatty acids was increased approximately 11.8% at 10oC compared to 30oC, although the proportion in the wild-type remained unchanged.
