The agitation speed and aeration rate for the production of carboxymethylcellulase (CMCase) by Cellulophaga lytica LBH-14 were established using one-factor-ar-a-time experiment and response surface methodology. The optimal agitation speed for cell growth of C. lytica LBH-14 under aeration rate of 1.0 vvm in a 7 L bioreactor (Ko-Biotech, Korea) was 400 rpm, which was the same as for production of CMCase and cellobiase. The optimal aeration rate for cell growth of growth of C. lytica LBH-14 under agitation speed of 400 rpm was 1.0 vvm, which was also the same as for production of CMCase and cellobiase. However, the optimal agitation speed and aeration rate extract by Design Expert software (Version 8.0, Stat-Ease Inc., USA) based on the response methodology were different from those for production of CMCase and cellobiase. The optimal agitation speed and aeration rate for cell growth were 500 rpm and 1.50 vvm, whereas those for productions of CMCase and cellobiase were 300 rpm and 0.50 vvm and 369 rpm and 0.93 vvm, respectively. More significant factor for cell growth was agitation speed, whereas that for production of CMCase and cellobiase was aeration rate.
