Decolorization of syntheric dyes with Bacillus subtilis spore

In this study, we tried to decompose synthetic industrial dyes using Bacillus subtilis spore, which has CotA spore coat protein known as having multi-copper oxidase (laccase) activity. Laccase from Trametes versicolor was used for control experiments. Bacillus subtilis spore was prepared using Schaeffers’ sporulation medium in shaking incubator after 5 day's culture at 37℃. Renografin was used for spore purification, and purifed spore was lyophilized and suspended in 0.1M sodium acetate buffer[pH5.0]. Synthetic dye such as Acid red 18(New coccine), Acid green 25, Indigo carmine were used for the experiments with varing temperature and pH conditions. And the effect of mediator addition was also examined.
