Post-translational modification (PTM) is a process in which biochemical functional groups are attached to specific amino acids. This may evoke structural changes in the protein, which is important to the function of the protein. To study the role of PTMs to the functions of proteins, especially in the protein-protein interactions, the non-proteinogenic, PTM-containing amino acids can be incorporated into a protein in a site-specific manner. To this end, we have shown that using CCCG four-nucleotide codon for genetic code reprogramming in the crude S12 system is the most reliable and robust method of choice (Ki Baek Lee et al., 2013). With the developed method, we present the PTM-specific protein-protein interactions using histone H3.1 and AIMP3/p18 as model proteins and lysine modifications (methylation and acetylation) as model PTMs.
