Using the mRNA differential display technique, the mRNA expression levels were detected after exposure to 10% (v/v) toluene for 8 h in a solvent-tolerant strain Pseudomonas sp. BCNU 106. Among the total of 100 up-expressed products, 51 cDNA fragments could be confirmed to clone and sequenced. Blast analysis revealed transcription, inorganic ion transport and metabolism, signal transduction and mechanisms, carbohydrate transport and metabolism, and DNA replication, recombination and repair, even defence mechanisms were found to display an adaptative activation to toluene. Furthermore, the RNA corresponding to ABC transportor, Mn2+and Fe2+ transporter, SOD, and cat genes was confirmed to be induced under the presence of 10% toluene by blot analysis. Thus, the cellular solute uptake and antioxidant defence system were essential for the tolerance to toluene in a solvent-tolerant strain Pseudomonas putida.
