Lipocalin-2(LCN2) is a secretory glycoprotein overexpressed from various inflammation and cancers. LCN2 was already known as a sensitive biomarker for renal cell carcinoma and chronic kidney diseases. Serum LCN2 was measured using a commercial ELISA kit, and other biochemical indicators including creatinine, leukocyte count, eGFR and C-reactive protein were estimated. Disadvantage of these ways based on immune reaction are time-consuming, high cost and low sensitivity. Recently, aptamers are gaining increasing interest as an alternative material of antibodies, because it is low cost, easy modification, high stability and specificity with target molecule. Therefore, we have developed a DNA aptamer based sensor for specific detection of LCN2. We performed the cloning and expression of LCN2 gene. We carried out the SELEX (Systematic Evolution of Ligands by EXponential enrichment) technique. After 10 cycles of SELEX, we selected the specific binding aptamers to LCN2 through Real-time PCR and Nanodrop spectrophotometer. Using the Surface Plasmon Resonance (SPR), aptamers obtained the high affinity and specific binding with LCN2. Aptamer-Linked Immobilized Sorbent Assay like an ELISA performed for quantitative measurement of LCN2. In conclusion, aptasensor based on specific LCN2 binding aptamers can be readily applied to rapid and sensitive detection of LCN2 from patients with cancer and noncancerous diseases.
