Recombinant antibodies are high value biomolecules which have been widely used for research, diagnostic and therapeutic purposes. Among these, single chain variable fragment (scFv), a fusion protein of the variable regions of the heavy and light chains of immunoglobulins, is the smallest active fragment and has several advantages over other antibody fragments, such as cheaper and faster production, higher penetration and clearance due to smaller molecular weight, etc. In recent years, C. glutamicum has been demonstrated as industrial protein factory because of its ideal attributes for production of proteins, such as secretion of few endogenous proteins, low level of extracellular proteolysis, capability of high cell density cultivation, etc. Here, we demonstrated C. glutamicum as potential recombinant antibody fragment manufacturer. Codon optimization of a scFv gene was introduced, and several expression systems with strong promoters and efficient 5' and 3' UTR sequences were examined. Also, fed-batch cultivation conditions were optimized for higher scFv productivity, and significantly improved productivity of scFv in extracellular medium could be achieved.
