In the present study, the effects of treadmill exercise on expression of myostatin and follistatin protein expression of old aged rats according to skeletal muscle types. 16-month-old male rats(n=12) and male Sprague-Dwaley (n=12) were used as the old-aged groups. The Sprague-Dawley rats were randomly divided into 4 groups(n=6 in each group), as follows: the old-aged female group(n=6), the old-aged female and exercise group(n=6), the old-aged male group(n=6), old-aged male and exercise group(n=6). The rats in the exercise groups were forced to run on a motorized treadmill for 30 min once a day for 4 weeks. The load of exercise consisted of running at a speed of 2 m/min for the first 5 min, 5 m/min for the next 25 min, with a 0° inclination. The expression of myostatin in soleus muscle was lower in the old aged male rats compared with old aged female rats. In contrast, the expression of follistatin in soleus muscle was higher in the old aged male rats compared to old aged female rats. Treadmill exercise decreased expression of myostatin and, increased expression of follistatin in the soleus muscle. The expression of myostatin in gastrocnemius muscle was lower in the old aged female rats compared with old aged male rats. In contrast, the expression of follistatin in gastrocnemius muscle was not statistically significant. Treadmill exercise decreased expression of myostatin, however, the expression of follistatin was increased by treadmill exercise in the gastrocnemius muscle. Here in this study, we showed that treadmill exercise is a very useful strategy for preventing loss of muscle by inhibiting expression of myostatin and enhancing expression of follistatin.
