High light inducible polypeptides (HLIPs) are revealed to prevent distinction of Synechocystis sp. caused by destruction of photosystem Ⅱ in high light condition. This decrease energy conversion efficiency of light energy in photon to chemical energy in biomass. The knock-out of whole HLIPs led to cell death, so partial regulation of HLIPs was used in this experiment to examine enhancement of photosynthetic efficiency. Two types of mutants were made by using megaprimerPCR method and genotypes of these mutants were identified by Southern blot. And we carried out following test to confirm phenotypes of these mutants. We used BG11 medium containing 0 or 0.1 mM of glucose in photoautotrophic or mixotrophic condition, respectively. The result showed relatively higher growth rate and chlorophyll concentration in both mutants than those in wild type Synechocystis sp.. Especially this difference is distinct in mixothrophic cultivation condition.
