Enhanced 2,3-butanediol production of recombinant Klebsiella pneumoniae by improving expression of related genes

2,3-Butanediol(2,3-BDO) is one of the major metabolite in Klebsiella pneumoniae KCTC2242, which is a important chemical with wide applications such as butadien and methyl ethyl keton. To enhance 2,3-BDO production, three genes-acetolactate synthase (budB), alphaacetolactate decarboxylase (budA) and butanediol dehydrogenase (budC)-were identified in genomic DNA of K. pneumoniae. With the goal of enhancing 2,3-BDO production, these genes were cloned into pUC18K expression vector containing kanamycin resistance gene to generate strains SGJSB01-07. All strains were incubated in flask experiments and compared 2,3-BDO productivity of each. Selected one strain that produce the large amount of 2,3-BDO was grown in a bioreactor using a batch and fed-batch fermentation. As a result, The maximum production of 2,3-BDO in recombinant K .pneumoniae achieved through fed-batch mode. This suggest that it strain is an useful for industrial application related 2,3-BDO.
